摘要
目的观察慢性摄入酒精大鼠模型肺实质的变化,探讨酒精与肺纤维化的相关性。方法雄性Wistar大鼠16只,随机分为模型组和对照组,各8只。模型组每天自由饮用10%酒精,对照组则自由饮水。16周后全部处死,分离完整的肺组织并取相应的标本,观察肺组织大体及超微结构改变。比较两组肺间质胶原沉积的差异,同时比较转化生长因子β1(TGF-β1)在肺组织中的表达情况。结果透射电镜显示模型组部分肺组织结构破坏,细胞间隔可见胶原纤维沉积。模型组胶原沉积程度增高(P<0.05);TGF-β1在肺组织中的表达增强(P<0.05)。结论慢性摄入酒精可以引起大鼠肺实质损伤并导致肺纤维化。
Objective To investigate the effect of chronic alcohol intake on rat lung tissue and the relationship between alcohol and pulmonary fibrosis.Methods 16 Wistar rats were randomly divided into the control group(n=8)and the model group(n=8).The control group was administrated with water and the model group was only administrated with 10% alcohol(V/V).All rats were killed 16 weeks later and their lung tissues were completely extracted.The structural change of ultraminiature lung tissues was observated by transmission electron microscope(TEM).The difference of the interstitial lung collagen deposition and the expression of TGF-β1 between the two groups was compared.Results In the model group,the lung tissues structure was partly destructed and there was collagen fiber deposition in cell compartment.Compared with the control group,the deposition of collagen in the alveolar interval and the expression of TGF-β1 increased more(P0.05)in the model group.Conclusion Chronic alcohol ingestion can lead to rat lung injury and pulmonary fibrosis.
