摘要
目的:分离并初步鉴定小鼠羊膜组织中干细胞样细胞。方法:采用胶原酶消化、微球形成法分离小鼠羊膜组织中的干细胞样细胞,并通过免疫细胞化学染色检测干细胞标志物Oct-3/4、C-kit及ALP在分离所得细胞球的表达情况。结果:从小鼠羊膜组织消化下来的细胞在低吸附培养板中生长7d后可形成细胞球,且该细胞球阳性表达干细胞标志物Oct-3/4、C-kit及ALP。结论:初步鉴定本实验分离到的细胞球具有干细胞特性,可为进一步研究小鼠羊膜来源干细胞打下坚实的基础。
Objective: To isolate and identify stem cell-like cells from amniotic membrane of ICR mouse.Methods: Amniotic membrane of ICR mouse was treated with collagenase and then cultured in low attachment multi-well plates.After 7 days of culture,the formed spheres were identified by immunocytochemistry staining with stem cell markers,including Oct-3/4,C-kit and ALP(Alkaline Phosphotase).Results:Cells isolated from mouse amniotic membrane could form spheres,which expressed positively stem cell markers including Oct-3/4,C-kit and ALP.Conclusions:Stem Cell-like cells can be isolated from mouse amnion membrane by the mentioned methods above,and it's expected to facilitate the further investigation on characterizing mouse amnion membrane-derived stem cells.
