摘要
目的:研究丁香酚在大鼠体内的组织分布规律。方法:大鼠ig给予丁香酚后,应用高效液相色谱(HPLC)法测定丁香酚在大鼠心、肝、脾、肺、肾、胃、大肠、小肠、脑、前列腺中的含量。色谱柱为AllsphereODS-2RP-18(250mm×4.6mm,5μm),流动相为甲醇:0.5%冰醋酸=63:37(V/V,pH3.80),检测波长为280nm,柱温为30℃,流速为0.8mL.min-1。结果:丁香酚在大鼠心、肝、脾、肺、肾、胃、大肠、小肠、脑、前列腺的线性回归方程分别为A=185.87c-0.3951(0.9972)、A=174.63c-0.1013(0.9992)、A=164.5c+0.0174(0.9975)、A=191.98c-0.6649(0.9912)、A=192.34c-0.0897(0.9878)、A=180.26c+0.1690(0.9983)、A=146.29c+0.1356(0.9993)、A=159.07c+0.2213(0.9991)、A=158.58c-0.0504(0.9987)、A=97.218c+1.6503(0.9335)。丁香酚组织样品匀浆液浓度在0.011~2.158μg.mL-1范围内与峰面积比值呈良好线性关系。最低检测浓度为0.011μg.mL-1。结论:大鼠ig给药后,较快的分配到各脏器组织中,分布范围广,其中以肠、脾中水平较高,脑中较低。
OBJECTIVE:To study the tissue distribution regularity of eugenol in rats.METHODS:HPLC method was adopted to determine the contents of eugenol in heart,liver,spleen,lung,kidney,stomach,small intestines,large intestines,brain,prostate,respectively after intragastrical administration of eugenol in rats.Allsphere ODS-2 RP-18 column was used with mobile phase consisted of methanol-0.5%acetic acid(63:37),(V:V,pH 3.80) at the flow rate of 0.8 mL·min-1.The detection wavelength was set at 280 nm,and column temperature was 30℃.RESULTS:The linear regression equations of eugenol in above tissue were A=185.87c-0.395 1(0.997 2),A=174.63c-0.101 3(0.999 2),A=164.5c+0.017 4(0.9975),A=191.98c-0.664 9(0.991 2),A=192.34c-0.089 7(0.987 8),A=180.26c+0.169 0(0.998 3),A=146.29c+0.135 6(0.999 3),A=159.07c+0.221 3(0.999 1),A=158.58C-0.050 4(0.998 7),A=97.218c+1.650 3(0.933 5),respectively.The linear range of eugenol in homogenate was 0.011~2.158 μg·mL-1 and the lowest detection limut of gentianine was 0.011 μg·mL-1.CONCLUSION:After intragastrical administration,it spread to each viscera and tissue rapidly and widely.The level of it is higher in intestines and spleen,and lower in brain among them.
