异丙肾上腺素诱导慢性心力衰竭大鼠模型的建立 被引量:25

Establishment of model of isoprenaline-induced chronic heart failure in rats

摘要 目的建立异丙肾上腺素(ISO)诱导SD大鼠慢性心力衰竭(CHF)模型,并对其进行评估。方法将SD雄性大鼠分为4组,ISO组每组20只均腹腔注射ISO各1次,剂量分别为2、2.5、3mg.kg-1.d-1,建立大鼠CHF模型,并用等量生理盐水制备对照组(n=10),分别在用药后4周进行心脏超声检测左室舒张末内径(LVEDD)、左室收缩末内径(LVESD)、左室射血分数(LVEF)、左室缩短分数(LVFS),4周末测血清B型钠尿肽(BNP)值、计算心脏质量指数。结果注射剂量为3.0mg.kg-1.d-1组4周末大鼠死亡14只,注射剂量为2.0mg.kg-1.d-1无大鼠死亡,2.5mg.kg-1.d-1组4周末大鼠死亡2只,大、中、小剂量组存活率分别为30%、90%及100%。ISO组LVEDD、LVESD、BNP及心脏质量指数均高于对照组(P<0.01),LVEF、FS低于对照组(P<0.01)。ISO组LVEDD、LVESD、BNP及心脏质量指数随着ISO剂量的增加而增大,组间比较差异有统计学意义(P<0.01),LVEF、FS随着ISO剂量的增加而降低,组间比较差异有统计学意义(P<0.05)。结论皮下注射ISO诱导建立大鼠CHF模型的方法简便、可靠;心衰模型的成功建立以ISO皮下注射剂量为2.5mg.kg-1.d-1较为适宜。 Objective To establish the model of isoprenaline(ISO)-induced chronic heart failure(CHF) in rats and to perform its evaluation.Methods 70 male SD rats were divided into four groups,normal group(n=10),2.0 mg·kg-1·d-1 group(n=20),2.5 mg·kg-1·d-1 group(n=20) and 3.0 mg·kg-1·d-1 group(n=20).HF was induced by subcutaneous injections of ISO with different doses for 4 weeks.Meanwhile,the control group was established by subcutaneous injecting same volume of normal saline.Four weeks later,the rat survival rates were compared,the cardiac function tests were performed by using Doppler echocardiography procedures and detected BNP,the heart weight/body weight ratio.Results The survival rates of high to low dose injection group were 30%,90% and 100%.Compared with control group,the LVEDD,LVESD,BNP and heart weight/body weight ratio were significantly increased(P0.01),LVEF and FS were decreased(P0.05).In the control group,the more ISO was injected,the higher were LVEDD,LVESD,BNP and heart weight/body weight ratio(P0.01),meanwhile the lower were LVEF,FS(P0.05).Conclusion The CHF model induced by subcutaneous injecting ISO is a simple and convenient method.Although the cardic function is much worse in 3 mg·kg-1·d-1,the survival rate is also lower than other groups.2.5 mg·kg-1·d-1ISO is a reliable dose for establishing the CHF model.
