摘要
目的研究TIMP-3基因在肝细胞肝癌(human hepatocellular carcinoma,HCC)中的表达,探讨其在HCC发生发展中的作用及意义。方法应用免疫组化(SP法)和SYBR Green实时荧光定量聚合酶链反应检测30例HCC及对应癌旁组织以及10例正常肝组织中的TIMP-3蛋白及TIMP-3mRNA的表达情况,分析TIMP-3基因的表达与HCC临床病理参数的关系。结果 TIMP-3蛋白明确定位于肝细胞胞质内。TIMP-3mRNA和TIMP-3蛋白在HCC中的表达量分别为明显低于癌旁肝组织和正常肝组织中的表达量,差异有统计学意义(P<0.01)。统计学分析显示,在HCC中TIMP-3基因的低表达与肿瘤分化程度、血清中甲胎蛋白表达水平、是否合并HBV感染等有关(P<0.05),与患者性别等因素无关。结论 TIMP-3基因可能在HCC的发生发展中起着重要作用,其表达有望成为HCC的重要分子生物学指标。
Objective To investigate the expression of TIMP - 3 gene in human hepatocellular carcinoma (HCC) and discussits significance during hepatocellular carcinogenesis. Methods Expression of TIMP - 3 protein in HCC and the paired paracarcinomaliver tissues from 30 HCC patients was detected by immunohistochemistry (SP method), and TIMP -3 mRNA expression was analyzed by reverse transcription polymerase chain reaction. The relationship between expression of TIMP - 3 geneand clinicopathologic parameters in patients with HCC was analysed. Results Expression of TIMP - 3 protein was definitely locatedin the liver cancer cell plasma. The positive rates of TIMP - 3 mRNA and TIMP - 3 protein in HCC were respeetively which were prominently lower than those in the paired paracarcinoma liver tissues ( P 〈 0. 01 ). Statistical analysis showed that TIMP - 3 in HCC gene expression and low degree of tumor differentiation, serum alpha - fetoprotein expression, such as the merger of HBV infection (P 〈 0. 05 ), gender and other factors unrelated to the patient. Conclusion TIMP - 3 gene may occur in the development of HCC plays an important role, its expression is expected to become important molecular targets of HCC.
