摘要
目的:研究蝎素组分Ⅲ(Scorpion venom crudeⅢ,SVC-Ⅲ)对THP1细胞HMGB1表达的影响,探讨蝎素与HMGB1相互作用的机制。方法:不同浓度SVC-Ⅲ(0.1、1.0、10和100 ng/ml)刺激培养THP1细胞48小时后,RT-PCR检测HMGB1 RNA水平表达差异;Western blot检测HMGB1蛋白水平表达情况;激光共聚焦检测HMGB1在细胞中的分布情况。结果:1.0 ng/ml SVC-Ⅲ刺激培养THP1细胞,HMGB1 RNA及蛋白表达水平升高最显著,随着SVC-Ⅲ浓度的增加,HMGB1 RNA及蛋白的表达水平反而下降;SVC-Ⅲ刺激培养THP1细胞可引起HMGB1从胞核转移入胞浆。结论:SVC-Ⅲ可以刺激或抑制THP1细胞HMGB1的表达,其作用效果与剂量密切相关。
Objective :'Researching the effect of SVC-Ⅲ on the expression of HMGB1 in THP1 cells in order to investigate the interactional mechanism of SVC- Ⅲ and HMGBI. Methods :THPI cells were stimulated by 0.1 ng/ml, 1.0 ng/ml, 10 ng/ml and 100 ng/ml SVC-Ⅲ for 48 hours,then detected HMGBI RNA level by RT-PCR,HMGB1 protein level by Western blot,distribution by laser scanning eonfocal. Results:HMGB1 RNA and protein expression were increased conspicuously by 1.0 ng/ml SVC-11I ,but dicreased a- long with the increasing Svc-m concentration. SVC-III can migrate HMGB1 from nucleus to cytoplasm. Conclusion :The effect of SVC- m on the expression of HMGB1 in THP1 cells have relation to the SVC-11I's concentration.
