摘要 目的探讨阿仑膦酸钠(ALN)对骨髓间充质干细胞(BMSCs)增殖和骨向分化能力的影响,观察其促BMSCs骨向分化过程中对转化生长因子β1(TGF-β1)及骨形成蛋白-2(BMP-2)表达的影响,分析可能的作用途径。方法以碱性磷酸酶(ALP)等指标确定干预药物不同浓度梯度中最佳促BMSCs骨向分化剂量;分组实验,通过检测诱导分化过程中ALP和钙化结节等指标的表达,观察其对BMSCs骨向分化能力的影响;ELISA法检测骨向分化过程中TGF-β1、BMP-2的表达。结果 ALN最佳促BMSCs增殖和骨向分化浓度为1×10-8mol/L,该浓度ALN能促进成骨指标的表达和TGF-β1和BMP-2分泌增加;具有促BMSCs骨向分化能力的ALN各诱导组均伴随TGF-β1和BMP-2分泌增加。结论 ALN能促进BMSCs增殖和骨向分化;在BMSCs骨向分化过程中,上调TGF-β1、BMP-2的表达量可能是各干预药物促进MSCs骨向分化的作用机制之一。 Objective To investigate the effects of alendronate(ALN) on the proliferation and differentiation of bone mesenchymal stem cells(BMSCs),and on the expressions of transforming growth factor-1(TGF-1) and bone morphogenetic protein-2(BMP-2) in the osteoblast differentiating progress of BMSCs.Methods The most effective concentration of ALN for promoting the osteoblast differentiation of BMSCs was confirmed according to some indexes(like alkaline phosphatase,ALP).A grouped experiment was conducted for the sake of studying the effect and mechanism of ALN in the process of inducing BMSCs differentiation into osteoblast through detecting expression of ALP and calcium nodes,as well as the expressions of TGF-β1 and BMP-2 with ELISA.Results The most effective concentration of the ALN on the BMSCs differentiation was 1×10-8mol/L,enhancing the expressions of osteoblast indices and the secretion of TGF-β1 and BMP-2.Besides,the increase of TGF-β1 and BMP-2 was revealed in all the groups,in which ALN showed its influence visibly.Conclusion ALN promotes the differentiation of BMSCs into osteoblast,and the up-regulation of TGF-β1 and BMP-2 possibly contributes to various interventional drugs in their differentiation promoting progress.
