组蛋白去乙酰化酶6在胃癌组织中的表达及意义 被引量:4

摘要 目的检测组蛋白去乙酰化酶6(HDAC6)在胃癌组织中的表达及其临床意义。方法采用免疫组化和原位杂交法检测74例胃癌及30例正常或黏膜慢性炎组织中HDAC6蛋白及mRNA的表达情况,并分析其表达与胃癌临床病理特征的关系。结果 HDAC6蛋白在胃癌组织中的阳性表达率(79.7%)显著高于正常或黏膜慢性炎组织(20.0%)(P<0.05),并且其在胃癌组织中的阳性表达率随浸润深度的增加而显著增高(阳性率分别为64.0%、77.8%、88.2%及100.0%,P<0.05);在有淋巴结转移者中HDAC6蛋白的阳性表达率(92.9%)显著高于无淋巴结转移者(62.5%)(P<0.05),其蛋白表达与癌的分化程度无关(P>0.05)。HDAC6 mRNA在胃癌组织中的阳性表达率(64.9%)显著高于正常或黏膜慢性炎组织(10.0%)(P<0.05),并且其在胃癌组织中的阳性表达率随浸润深度的增加而显著增高(阳性率分别为48.0%、55.6%、76.5%及92.9%,P<0.05);在有淋巴结转移者中HDAC6 mRNA的阳性表达率(78.6%)显著高于无淋巴结转移者(46.9%)(P<0.05),其mRNA表达与癌的分化程度无关(P>0.05)。HDAC6蛋白与mRNA的表达存在相关性(C=0.374,P<0.01)。结论 HDAC6的过表达可能与胃癌的发生发展有关,并对判断胃癌的生物学行为具有一定的意义。
