珍稀藏药桃儿七组织培养研究 被引量:10

摘要 建立以带胚种段为外植体的藏药桃儿七(Sinopodophyllum hexandrum)组织快繁培养体系,研究了外植体不同处理方法,不同激素配比对无菌苗诱导的影响;活性炭(AC)、维生素C(Vc)和聚乙烯吡咯烷酮(PVP)对无菌苗褐变的抑制效果。结果表明:带胚种段经诱导、增殖、生根系列培养基培养后能获得长势良好诱导率较高的无菌苗,诱导率为90.0%。胚诱导培养基为MS+20 g/Lsucrose+1 g/L AC;诱导培养基为MS+0.5 mg/LIAA+0.5 mg/L KT+1.0 mg/L GA3+20 g/Lsucrose+1 g/L PVP;增殖培养基为MS+2.0 mg/L 6-BA+0.3 mg/LGA3+20 g/L sucrose+1 g/L PVP;生根培养基为MS+0.5 mg/L IAA+0.5 mg/L KT+0.5 mg/L GA3+20 g/L sucrose+1 g/L PVP。 The tissue culture system of explants from separated embryo seeds of matured Tibetan medicine Sinopodophyllum was established.The effects of seedling induced rate by different treatments and different concentrations of hormones,activated charcoal(AC),vitamin C(Vc) and polyvinyl pyrrolidone(PVP) were studied.The results indicated that seedling induced rate could reach 90.0% with better growth and development on the series of media including induction,proliferation and rooting.The embryo induction medium was a base of MS+1 g/L AC;the induction medium was MS+0.5 mg/L IAA+0.5 mg/L KT+1.0 mg/L GA3+1 g/L PVP;proliferation media was MS+2.0 mg/L 6-BA+0.3 mg/L GA3 +1 g/L PVP and the rooting medium was MS+0.5 mg/L IAA+0.5 mg/L KT+0.5 mg/L GA3+1 g/L PVP.In addition,2 g/L sucrose and 0.4% agar were the common components of the four.
