摘要
目的研究脊髓损伤急性期对成年大鼠睾丸生精功能障碍的影响及其机制。方法80只SD雄性大鼠随机分为脊髓损伤组(40只)和脊髓非损伤组(40只),采用原位细胞凋亡TUNEL法检测脊髓损伤后第1、2、4、8、16天睾丸生精细胞的形态及数量变化。4、8、16天睾丸生精细胞Fas、Bcl-2表达情况。结果采用免疫组化SABC法检测大鼠脊髓损伤后第1、2、大鼠脊髓损伤后睾丸TUNEL标记阳性凋亡细胞第4天开始出现,多表达于初级、次级精母细胞及精原细胞,至第16d,随时间点推移阳性凋亡细胞数逐渐增多。脊髓损伤后第4~16d损伤组睾丸生精细胞Fas表达与非损伤组比较显著增强(P〈0.05),Bcl-2表达损伤组与非损伤组差别无显著性(P〉0.05)。结论大鼠脊髓损伤急性期存在睾丸生精细胞凋亡现象,睾丸生精功能障碍与Fas高表达密切相关,和Bcl-2表达无明显相关性。
Objective To study the cause of abnormalities in spermatogenesis of the male rats after spinal cord injury. Methods Eighty sexually matured Sprague-Dawley rats were randomly divided into two groups: T9 surgical transection and sham control. Immunohistochemical SABC was used to detect the expression of Fas and Bcl-2 protein in the T9 surgical transaction and sham-operation rats on the 1st, 2nd, 4th, 8th and 16th days after surgery. Terminal deoxynuleotidyl-mediated nick end labeling (TUNEL) was used to detect the expression of spermatogenic cell apoptosis after surgery (1s', 2nd, 4th, 8th, and 16th days). Results After injury, Fas expression of the surgical group increased significantly (P〈0.05) on the 4th, 8th,,and 16th days compared with the sham control group, but there was no difference in Bcl-2 expression between the surgical group and the sham control group (P〉0.05). TUNEL-positive cells were found mostly in the spermatocytes and spermatogonia on the 4th day. after surgery, and from the 4th-16th days more and more positive cells were found. Conclusion There is apoptosis of spermatogenic cells after spinal cord injury, and high expression of Fas is one of the reasons causing abnormalities in spermatogenesis. But it has nothing to do with Bcl-2 expression.
