摘要
以表雄酮为原料,采用固体酸催化剂,一步脱水合成肌肉松弛药的中间体5α-雄甾-2-烯-17-酮。通过对反应温度、催化剂和表雄酮的配比及催化剂的酸度进行优化,得出优化工艺条件:反应温度105℃,m(催化剂)∶m(表雄酮)=5∶1,催化剂酸度为4.5%,反应时间2 h,收率87.1%。
5α-Androst-2-en-17-one which is an intermediate of muscle relaxants was synthesized from commercially available epiandrosterone catalyzed by silica supported sulfuric acid.Influences of the reaction temperature,the ratio of m(catalyst)∶m(epiandrosterone) and the acidity of the catalyst were investigated.The optimum reaction conditions were obtained as follows: reaction temperature 105℃,m(catalyst)∶m(epiandrosterone)ratio 5:1,catalyst acidity 4.5%,reaction time 120 min.The product was resulted in 87.1% yield.
