成年态南丰蜜橘离体再生芽培养及芽维持的蛋白谱特征 被引量:1

摘要 以成年态南丰蜜橘茎段为材料,在不同激素配比的MS培养基中进行再生芽的诱导和继代增殖培养,确定了最佳的诱导培养基改良MS+6-BA0.5 mg/L+KT0.5 mg/L+NAA0.1 mg/L,把诱导培养基中的蔗糖浓度由3%提高到6%可以很好地解决再生芽的叶片发黄、脱落问题;再生芽增殖的最佳培养基为改良MS+6-BA0.4 mg/L+ZT0.1 mg/L+NAA0.05 mg/L,增殖系数达5.40.SDS-PAGE电泳结果表明33.4 kD相对分子质量的蛋白谱带出现于再生芽正常分化生长时期,可能是维持再生芽生长发育的特异蛋白组分. In this paper, the mature stem segments of Citrus reticulita Blanco were used as explants for buds induction and subculture culture on medium with different hormone combinations .The results showed that the best induction medium is improvement MS+6-BA0.5 mg/L +KT0.5 mg/L + NAA0.1 mg/L. The induction medium of sucrose concentration from 3% to 6% can solve the leaves of buds yellow, shedding problems; subculture proliferation medium of buds is improvement MS+6-BA0.4 mg/L+ZT0.1 mg/L+NAA0.05 mg/L. Proliferation of multiple was 5.40. The results of SDS-PAGE show that 33.4kD protein is only expression in the period of the buds begin to normal growth. It probably the specific protein of buds maintained for normal growth.
