摘要
目的 探讨链脲佐菌素诱发的糖尿病模型大鼠的胰腺组织中自由基清除剂 SOD m RNA和磺脲类药物受体SUR m RNA的表达 ,了解糖尿病时胰腺 SOD和 SUR的关系。方法 分别在 0、4、12、2 4周取其血和胰腺 β细胞测定血糖、血脂、SOD、SUR的 m RNA表达研究 [采用逆转录 聚合酶链反应 (RT PCR) ]。结果 和正常对照组相比 ,糖尿病大鼠血糖、血脂缓慢上升 ,胰腺 β细胞的 SOD m RNA表达量明显低下 ,SUR的 m RNA也有类似改变 ,但晚于 SOD的变化 ,相关分析示二者密切相关 ,r=0 96,P<0 0 0 1。结论 2型糖尿病模型大鼠的胰腺 β细胞中存在 SOD、SUR的 m R NA表达水平低下 ,而且随着高血糖持续时间的延长 ,呈逐渐下降趋势。胰腺组织中 SOD改变可能是糖尿病时 SUR水平下降的原因之一。
Objective Probing into the expression of scavenger SOD and SUR's mRNA in pancreatic tissue of diabetic rats induced by streptozocin to understand relationships between SOD and SUR.Methods We quantified the blood sugar, lipoprotein and the gene expression of SOD and SUR by RT PCR in beta cells of pancreatic tissue at 0, 4, 12, 24 weeks separately.Results Compared with control group, the levels of blood sugar and lipoprotein increased slowly; the levels of expression of SOD and SUR mRNA were lower in beta cells of pancreatic tissue. The relationship between SOD and SUR mRNA was close.Conclusion The results indicated that there was a low expression of SOD and SUR gene in beta cells of pancreatic induced by streptozocin, which become distinct especially during the longer periods of hyperglucose. The changes of SOD mRNA expression in pancreatic tissue may be one of reasons resulting in lowing SUR mRNA levels.
