摘要 [目的]研究聚半乳糖醛酸酶高产菌株的复合诱变条件。[方法]黑曲霉孢子经紫外线辐射2 min后,涂布于含0.8%LiCl的培养基上培养,经初筛和复筛后测定聚半乳糖醛酸酶活力。[结果]获得1株产聚半乳糖醛酸酶活力为10.450万U/ml的菌株,产酶活力比出发菌株提高了60.77%。[结论]产聚半乳糖醛酸酶的黑曲霉经紫外线辐射2 min、0.8%LiCl复合诱变后,产酶活力提高了60.77%。 [Objective]The aim was to study the compound mutation conditions of high producing strains yielding PG.[Method] After radiating for 2 min,Aspergillus niger spores are spread on culture medium with 0.8% LiCl,then after first screening and second screening,enzyme activity of PG are measured.[Result] A bacterial strain with 10.450×104 U/ml enzyme activity of PG are obtained,its enzyme activity of PG is improved by 60.77% than original strains.[Conclusion] After radiating for 2 min,Aspergillus niger spores are spread on culture medium with 0.8% LiCl,its enzyme activity of PG is improved by 60.77%.
