广西南部地区甘蔗黄叶病毒病的研究 被引量:5

摘要 [目的]明确甘蔗黄叶病毒病SCYLV在广西南部蔗区的发生情况,为广西健康种苗的推广和抗病育种提供科学依据。[方法]在广西南部蔗区对不同甘蔗品种采集显症或不显症甘蔗叶片,采用引物(P1:5'-aatcagtgcacacatccgag-3'/P2:5'-ggagcgtcgcctacctatt-3')进行RT-PCR检测。[结果]对经P1、P2扩增获得的PCR产物进行直接测序,所得序列在NCBI中比对确认均为SCYLV CP序列。黄叶病调查结果发现,钦州蔗区感病最严重,品种感病率达80%;扶绥蔗区的5个品种中,检出黄叶病毒的有3个品种,感病率为60%;凭祥蔗区感病的品种占60%;北海蔗区的品种感病率为50%;宁明东安乡蔗区6个品种中1个品种检出SCYLV,感病率为17%;驮卢镇渠立村和崇左江州2个地方11个品种均未检测出SCYLV。[结论]甘蔗黄叶病毒病已在广西南部大部分蔗区发生且蔓延。 [Objective] In order to determine the occurrence of sugarcane yellow leaf virus(SCYLV)in the southern region of Guangxi,to provide a scientific basis evident to disease-free sugarcane producing and resistant disease breeding in Guangxi.[Method] Sugarcane leaf samples with symptom or symptomless were collected from commercial growing fields in the southern of Guangxi.The samples were detected by RT-PCR with SCYLV specific primers(P1:5′-aatcagtgcacacatccgag-3′/P2:5′-ggagcgtcgcctacctatt-3′).[Result] The amplified cDNA products were sent to sequence directly.BLAST analysis revealed that they all were high homolog to reported SCYLV CP gene.The investigation results showed that the percentage of infected varieties was 80% in Qinzhou,being the highest infection area in southern region of Guangxi.There were high SCYLV infected varieties rates in another three towns included Fusui,Pingxiang,Beihai,which were 60%,60% and 50%,respectively.One variety was found to be positive by detection with RT-PCR from 6 varieties in Ningming Dongan Village,which hold 17%,while no variety was detected to be positive from 11 varieties in Tuolu and Jiangzhou Chongzuo.[Conclusion] The virus has occurred and spread in most of sugarcane growing areas in southern Guangxi.
