摘要
[目的]分离筛选出能够降解土壤中多菌灵的菌株,并对其降解特性作初步研究。[方法]从长期被多菌灵污染的土壤中,利用富集培养法分离筛选出1株能够降解多菌灵的菌株YB-6,并研究了初始pH、培养温度、接种量、外加碳源、氮源对其生长量和降解特性的影响。[结果]菌株YB-6能够以多菌灵为唯一碳源生长,在基础培养基中,5 d内对100 mg/L的多菌灵降解率为61%,而加入0.5%蛋白胨后5,d内对多菌灵的降解率可达90%;菌株BY-6对多菌灵具有良好降解性能,最合适降解温度为30℃、pH为7.0,降解率与接种量呈正相关,外加氮、碳源能促进其降解效果。[结论]研究结果为进一步研究菌株YB-6降解多菌灵机理,并将其大规模运用到土壤修复技术中奠定了基础。
[Objective] To isolate bacterium strains which could degrading carbendazim in soil and study the biodegradation characteristics of bacterium strains.[Method] A bacterium YB-6 was screened and isolated from the carbendazim polluted soil.The effect of some factors on its biodegradation characteristics in vitro were studied,such as initial pH,culture temperature,inoculums size,extra carbon source,nitrogen source and initial concentration of carbendazim.[Result] The results showed that bacterial strain YB-6 could grow by using carbendazim as the sole carbon source.It could degrade 55% of 100 mg/L carbendazim in mineral culture medium in 5 d,but could degrade 90% by adding 0.5% peptone as nitrogen source.The degradation property of carbendazim by BY-6 is good and the optimal pH and culture temperature for biodegradation are 7.0 and 30 ℃ respectively.The biodegradation speed was related positively to initial inoculums size.
