摘要
目的 研究药物作用前后人高转移肺癌细胞中转移相关基因表达的改变,进一步揭示淫羊藿甙(ICA)及济南假单胞菌制剂PJA 抗转移作用的机制。方法 流式细胞术及逆转录PCR(RT-PCR)技术检测转移相关基因c-m yc、Tiam -1、nm 23-H1 蛋白及m RNA 水平的变化。结果 经PJA 及ICA 处理48 h 后,PG 细胞 c-m yc、Tiam -1 基因m RNA均有不同程度地降低,而nm 23-H1 m RNA水平有不同程度地升高,药物处理96 h 后,基因水平的变化已反映至其表达产物-蛋白水平上。c-m yc及nm 23-H1 基因蛋白的升降趋势与其m RNA 水平相一致(P< 0.005),且两种水平的变化均以协同作用组最显著。结论 PJA、ICA 通过抑制转移诱导基因c-m yc、Tiam -1的表达,促进转移抑制基因nm
Objective To investigate the change of expression of metastatic correlated genes in human highly metastatic lung tumor cell line and to explore the mechanism of anti metastasis by icariin(ICA) and pseudomonas jinanensis agent (PJA).Methods In this study flow cytometry and RT PCR technique were used to detect the change of protein and mRNA levels of c myc,Tiam 1 and nm23 H1 genes.Results After treated with PJA or ICA for 48 hrs,the relative expression levels of both c myc and Tiam 1 mRNA decreased in compare with that of the control.On the contrary,an enhanced tendency in the expression level of nm23 H1 mRNA was observed.After treated with the combination of two drugs for 96 hrs, changes of gene expression were detected on protein level.Change tendency of c myc and nm23 H1 gene oncoprotein expression levels was in accordance with that of mRNA levels,and the effect of combination group was most significant.Conclusion These data suggested that PJA and ICA inhibited the expression of metastatic induced genes c myc、Tiam 1 and enhanced the expression of metastasis suppressor gene nm23, and thus could be the anti metastatic mechanism of PJA and ICA.$$$$
