摘要
[目的]分离纯化北五味子茎藤中具有抗肝癌细胞活性的化学成分.[方法]反复采用硅胶柱色谱及重结晶等方法分离化合物.根据理化性质和波谱学分析,鉴定化学结构.并采用MTT法分析化合物的体外抗肝癌细胞作用.[结果]分离得到7个化合物,分别鉴定为五味子丙素(Ⅰ)、十九醇(Ⅱ)、三十五醇(Ⅲ)、二十四醇(Ⅳ)、五味子乙素(Ⅴ)、β-谷甾醇(Ⅵ)及五味子醇乙(Ⅶ).活性检测结果见,化合物Ⅰ,Ⅴ,Ⅵ有较强的体外抗肝癌细胞活性,质量浓度均为50mg/L时的抑制率分别为96.27%,97.11%,93.99%;化合物Ⅱ,Ⅲ,Ⅳ,Ⅶ的质量浓度均为50mg/L时抑制率分别为86.81%,82.33%,77.62%,86.25%.[结论]从北五味子茎藤中分离得到的7个化合物均具有体外抗肝癌细胞活性.
OBJECTIVE To isolate and purify the active constituents of the Schisandra vine stem anti-hepatocellular carcinoma cells.METHODS The components were isolated repeatedly from Schisandra vine stem by using silica gel column and recrystallization,and their structures were determined by means of physicochemical properties and spectral analysis,and the effects of anti-hepatocellular carcinoma cells in active components of these compounds were evaluated in vitro by MTT.RESULTS Seven components were identified as Schisandrin C(Ⅰ),19 alcohol(Ⅱ),35 alcohol(Ⅲ),24 alcohol(Ⅳ),γ-schiza-ndrin(Ⅴ),β-sitosterol(Ⅵ) and gomisin A(Ⅶ),and it was showed at mass concentration as 50 mg/L by activity test that the compoundⅠ,Ⅴ and Ⅵ had strong activity for anti-hepatocellular carcinoma cells in vitro and inhibitory rates were 96.27%,97.11% and 93.99%,and the compound Ⅱ,Ⅲ,Ⅳ and Ⅶ were 86.81%,82.33%,77.62% and 86.25%.CONCLUSION All seven compounds isolated form the Schisandra vine stem have activity for anti-hepatocellular carcinoma cells in vitro.
