大肠埃希菌提取物和LPS对单核巨噬细胞增殖与凋亡的影响 被引量:1

摘要 目的探讨不同浓度大肠埃希菌提取物(菌胞质蛋白)对单核细胞增殖和凋亡的影响。方法采用MTT法测定不同浓度的胞质蛋白和LPS作用72 h后,对U937细胞的细胞抑制率,并用流式细胞术检测胞质蛋白作用24 h U937细胞凋亡率。结果大肠埃希菌胞质蛋白对U937细胞增殖有显著抑制作用,并可诱导细胞凋亡。低浓度的LPS不能抑制U937细胞增殖,高浓度LPS可以明显抑制U937细胞增殖。结论大肠埃希菌提取物对单核巨噬细胞有显著的增殖抑制和诱导凋亡作用。 Objective To investigate the effect of Escherichia coli extract which main component is cytoplasm pro- teins on proliferation and apoptosis of U937. Methods The proliferation inhibition of cytoplasm proteins and LPS on U937 cells was determined by MTT reduction assay. Apoptosis rate of U937 were measured by flow cytometry after being treated with cytoplasm proteins for 24 hours. Results Cytoplasm proteins had the effect of inhibiting U937 cells proliferation and inducing them apoptosis. LPS could inhibited cells proliferation only in high concentration. Conclusion The cytoplasm proteins from Escherichia coli show strong potential for inhibiting U937 proliferation and inducing apoptosis.
