摘要
目的:建立反相高效液相色谱法测定血清中氨甲喋呤浓度的方法。方法:以甲醇∶醋酸盐缓冲液为流动相,茶碱作内标,检测波长为306nm。结果:在0~8μg·ml-1范围内呈良好线性关系,r=0.9999。日内及日间测得RSD分别为3.66%,4.03%。平均回收率为95.7%,血清中最低检测浓度为50ng·ml-1。结论:本法具有快速、准确,操作简便,流动相成本低等优点。
Aim To establish a HPLC method for the Determination of MTX in human serum.Methods Theophyline was selected as internal standard.The UV detector was set at 306 nm.The mobil phase consisted of methanol and 0.05 M HAC-NaAC buffur (30:100).Results the detection limit was 50ng·ml -1 .The good linearity was from 0 to 8 (g·ml -1 .The average recovery was 95.7%.Within day and between days were 3.66% and 4.03% respectly. Conclusion this method is sensitive,rapid accurate,inexpensive enough to be used for clinical therapeutic drug monitoring.
