E-钙黏蛋白(E-cadherin)对人滋养细胞侵袭能力及妊娠结局的影响 被引量:7

Effect of Adhesion Molecule E-cadherin on the Invasiveness of Human Trophoblast Cells and Outcome of Pregnancy

摘要 目的:探讨黏附分子E-钙黏蛋白(E-cadherin)的表达对人滋养细胞侵袭能力的调控及其与自然流产发生的关系。方法:分别以过表达或小干扰RNA(siRNA)的方法上调或下调人绒毛膜癌细胞株Bewo及JEG-3内源性E-钙黏蛋白表达,Transwell侵袭实验观察细胞侵袭行为的改变,并应用Western blotting比较4例正常妊娠妇女及4例自然流产妇女绒毛组织内E-钙黏蛋白的表达水平。结果:转染野生型E-钙黏蛋白质粒的Bewo及JEG-3细胞随着E-钙黏蛋白表达的提高,细胞的侵袭能力显著下降(P<0.05);而转染E-钙黏蛋白siRNA的Bewo及JEG-3细胞,随着E-钙黏蛋白表达的下降,细胞侵袭能力显著提高(P<0.05)。自然流产妇女较正常妊娠妇女的绒毛组织E-钙黏蛋白的表达水平不同程度出现升高。结论:E-钙黏蛋白参与调控滋养细胞的侵袭能力,其表达水平可能与妊娠结局密切相关。 Objective: To investigate the regulation of adhesion molecule E-cadherin expression on the invasiveness of human trophoblast cells and its relationship to the occurrence of natural abortion.Methods: The endogenous E-cadherin expression was increased or decreased in human choriocarcinoma cell line JEG-3 and Bewo by wild type E-cadherin plasmid transfection or small interference RNA(siRNA) transfection,and then the invasiveness of human trophoblast cells was detected using Transwell assay.Western blotting was then used to compare the difference in E-cadherin expression between villi obtained from 4 normal pregnancy women and that from 4 natural abortion women.Results: The invasiveness of Bewo and JEG-3 cells transfected with wild type E-cadherin plasmid significantly decreased(P0.05) accompanied with the increase of endogenous Ecadherin expression.The invasiveness of Bewo and JEG-3 cells transfected with E-cadherin small interfering RNA significantly increased(P0.05) accompanied with the decrease of endogenous E-cadherin expression.Ecadherin expression in villi obtained from natural abortion women was higher than that from normal pregnancy women.Conclusion: E-cadherin plays a key role in the regulation of invasiveness of human trophoblast cells,and its expression level may correlate with the outcome of pregnancy.
