摘要
[目的]对白皮松(Pinus bungeana)松针总RNA提取方法做比较,以得到完整的、高质量的白皮松总RNA。[方法]采用Trizol法、RNA提取试剂盒法和LiC l沉淀法3种方法提取白皮松总RNA,通过琼脂糖凝胶电泳和紫外分光光度计检测总RNA的完整性和提取纯度。[结果]LiC l沉淀法提取的RNA收率高,完整性好,OD260/OD280均在1.80~2.00,28S、18S条带清晰。[结论]LiC l沉淀法能高效地从白皮松针叶组织中分离出纯度高、完整性好的RNA样品。
[Objective] The aim of this study was to compare three RNA extraction methods and thus find out one suitable for isolating intact and high quality total RNA from Pinus bungeana.[Method] Employing three extraction methods of Trizol,RNeasy Mini Kit,LiCl precipitation,total RNAs of P.bungeana were extracted from pine needles samples,and their integrity and purity were detected via agarose gel electrophoresis and spectro photometry for a comparative study.[Result] Of the three extraction methods,LiCl precipitation method performed higher yield and better integrity of total RNAs extracted,with OD260/OD280 ratio between 1.8-2.0 and clear 28S and 18S bands in electrophoresis pattern.[Conclusion] LiCl precipitation method could be used to extract high pure and intact total RNA from P.bungeana.
