摘要 [目的]为了筛选出一种较准确地快速测定火龙果花粉生活力的方法。[方法]以红肉和白肉火龙果为试材,对其开花动态进行观察,并采用形态测定、碘-碘化钾染色、联苯胺染色、葡萄糖液培养和蔗糖液培养等5种方法对新鲜花粉和室内常温保存24 h的花粉进行生活力的测定。[结果]晚上21:30~22:30火龙果花瓣完全打开,花粉自然散开,是花粉采集的最佳时间;形态观测较适合用于火龙果花粉生活力的检测,碘-碘化钾染色法不适合,联苯胺染色法染色结果不准确,葡萄糖液培养和蔗糖液培养法检测花粉萌芽不理想。[结论]该研究为火龙果授粉、花粉采集及贮藏提供理论基础。 [Objective]The research aimed to screen out a fast and rapid detection method for the vitality of the pollen in the pitaya.[Method]Hylocereus polyrhizus(Weber) Britt. Rose(red pulp variety) and Hylocereus undatus(Haw) Britt. Rose(white pulp variety) were used as experimental materials to observe its flowering dynamics on detecting the vitality of fresh pollens and preserved pollens kept on indoor temperatures for 24 hours.5 detectinon methods involved morphometry method,I2-KI coloration method,benzidine coloration method,glucose solution culture method and sucrose solution culture method.[Result]The time 21:30-22:30 at night was the right moment for pollens collection as petals were fully bursting and pollens were dispersing.Morphometry method was the best method for testing vitality of fresh pollens while I2-KI coloration method wasn't fit for and benzidine coloration method's outcome was imprecise and glucose solution culture method and sucrose solution culture method wasn't good.[Conclusion]The research could provide the reference for the pollination,the pollens collection and the preservation.
