摘要
[目的]研究海洋硅藻叶绿素a测定方法的改进。[方法]通过对现有的叶绿素a测定方法(规范法、浸泡法、冻融法)进行比较分析,结合硅藻细胞自身特征,对海洋硅藻叶绿素a测定方法进行改进。[结果]将原先的冻融法和浸泡法中的手动振荡改为漩涡振荡20m in,并将浸泡法的浸泡时间减少为6 h。对比分析表明,改进后的冻融法和浸泡法的相对标准偏差均低于5%,较原先的冻融法和浸泡法有进一步的降低,且叶绿素a相对值较高。[结论]改进后的冻融法和浸泡法更加适合海洋硅藻叶绿素a含量的测定,具有很好的重复性,测量结果相对精确度高,操作简便,适用于数据量较大的试验。
[Objective]The research aimed to study the improvement of chlorophyll a measuring method in the marine diatom.[Method]The existing chlorophyll a measuring methods which included the criterion method,soaking method and freeze-thaw method were contrasted and analyzed.By combining with the self characteristic of diatom cell,the measuring method of chlorophyll a in the marine diatom was improved.[Result]The manual oscillation in the original freeze-thaw method and soaking method was changed into the vortex oscillation 20 min.Moreover,the soaking time in the soaking method decreased to 6 h.The contrast and analysis showed that RSD of improved freeze-thaw method and soaking method was lower than 5%,which had the further decrease than that of original freeze-thaw method and soaking method.Moreover,the relative value of chlorophyll a was higher.[Conclusion]The improved freeze-thaw method and soaking method were more suitable for the measurement of chlorophyll a content in the marine diatom.They had the good repeatability.The relative accuracy of measured result was high,and the operation was simple.They were suitable for the experiment whose data was large.
