酪蛋白激酶1基因在恶性黑色素瘤中的表达及临床意义 被引量:2

摘要 目的研究酪蛋白激酶-1(CK1)在恶性黑色素瘤细胞及组织中的表达并探讨其临床意义。方法采用qRT-PCR检测CK1 mRNA在恶性黑色素瘤细胞及正常黑色素细胞中的表达。Western blot检测CK1蛋白在恶性黑色素瘤细胞及正常黑色素细胞中的表达。应用免疫组织化学方法检测59例恶性黑色素瘤及30例色素痣标本中CK1的表达。结果恶性黑色素瘤细胞及组织中CK1基因表达明显高于在黑色素细胞及色素痣组织中的表达,差异有统计学意义(P<0.05)。结论 CK1基因在恶性黑色素瘤细胞及组织中高表达揭示CK1可能与恶性黑色素瘤的发病机制相关。以CK1为靶点的抑制剂或反义基因治疗黑色素瘤可能会展示广阔的前景。 Objective To investigate the expression of casein kinase-1(CK1) in malignant melanoma cells and tissues and its clinical significance. Methods CK1 mRNA was detected in malignant melanoma cells and melanocyte using qRT-PCR.Western blot was used to examine the expression of CK1 protein in malignant melanoma cells and melanocyte.Immunohistochemical stainings were used to examine the expression of CK1 protein in 59 melanoma tissues and 30 nevus tissues. Results The expression of CK1 gene was significantly greater in malignant melanoma cell lines and tissues than that in melanocyte and nevus tissues(P〈0.05). Conclusions The over-expression of CK1 gene in malignant melanoma cell lines and tissues might be related with pathogenesis of malignant melanoma.CK1 inhibitors and anti-sense gene therapy might be widely used in the future.
