脑微血管内皮细胞与体外培养神经干细胞共培养后Th1/Th2相关细胞因子的表达

The expressions of Th1/Th2 related cytokines after co-cultured microvascular en dothelial cells with neural stem cells in vitro

摘要 目的体外观察脑微血管内皮细胞(MVECs)对来自大鼠海马神经干细胞(NSCs)免疫相关细胞因子表达的影响。方法采用Tran-swell建立神经干细胞与脑微血管内皮细胞共培养模型(NSCs+MVECs),用荧光免疫化学和RT-PCR测定NSCs中与Th1相关的IFN-γ、IL-2及与Th2相关的IL-4、IL-10的表达。结果两种细胞共培养后,神经干细胞呈INF-γ,IL-2,IL-4,IL-10阳性;同时INF-γ、IL-2 mRNA的表达上调,而IL-4、IL-10mRNA的表达下调,与空白对照组比较明显差异(P<0.01)。结论脑微血管内皮细胞(MVECs)有可能使神经干细胞的INF-γ、IL-2表达上调,IL-4、IL-10的表达下调。 Objective To investigate microvascular endo thelial cells(MVECs)on the expressions of IFN-γ,IL-2,IL-4,IL-10 of NSCs in vitro. Methods Rat neural stem cells(NSCs) aged 3~5 day were ado pted for amplification in vitro,and co-cultured microvascular endothelial cell s with neural stem cells.After 48 h,the expressions of IFN-γ,IL-2,IL-4, IL-10 were assayed by immunohistochemistry and RT-PCR. Results After 48 h of co-cultured,the NSCs was positiv e of expressions in cytokine s INF-γ,IL-2,IL-4,IL-10;and the expressions of INF-γ,IL-2 were incre ased,the expressions of IL-4,IL-10 were deceased(P〈0.01). Conclusions MVECs mayb e up-regulate the expressions of IFN-γ,IL-2 and down-regulate the express ions of IL-4,IL-10 of NSCs.
