摘要
目的探讨虾青素对脂质代谢的调节作用,观察其对高脂诱发的主动脉及肝脏脂质过氧化损伤的影响。方法 50只鹌鹑随机分为5组,对照组给予普通饲料,其余4组给予高脂饲料;喂养同时虾青素低、高2剂量组分别以5、10mg/kg给予灌胃虾青素,凯-辛联用组按10mg/kg凯西莱,1mg/kg辛伐他汀灌胃。实验结束时取空腹血测定血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及丙二醛(MDA)含量、血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)及诱导型一氧化氮合酶(iNOS)活性;检测肝脏中TC、TG及SOD、GSH-Px及诱导型iNOS活性、MDA及还原型谷胱甘肽(GSH)含量;HE染色观察主动脉血管病理变化。结果与高脂模型组比较,虾青素低、高2剂量组及凯-辛联用组血清TG、TC、LDL-C含量及肝中TG及TC含量明显降低(P<0.05或<0.01),而血清HDL-C含量明显增高(P<0.01);血清SOD活性、GSH-Px活性及GSH含量明显提高(P<0.05或<0.01),而血清iNOS活性、MDA含量降低(P<0.05或<0.01)。结论虾青素具有良好的降脂功效,并对高脂诱发的主动脉及肝脏脂质过氧化损伤有明显抑制作用,高剂量虾青素改善效果更理想。
Objective To explore the regulatory effects of astaxanthin on lipid metabolism in quails and its influence on aorta and liver lipid peroxidation injury induced by high lipid. Methods 50 quails were randomly divided into 5 groups, the quails in control group were fed with common diet, however, the quails in the others were fed with high fat diet, at the same time, the quails in low dose and high dose astaxanthin groups were given astaxanthin by gavage at a dose of 5 mg/kg and 10 mg/kg respectively, and the quails in combined intervention group were given tiopronin ( 10 mg/kg) and simvastatin ( 1 mg/kg). At the end of experiment,the fasting blood of the quails were taken out, and the levels of total cholesterol ( TC ), triglyceride ( TG ), high density lipoprotein cholesterol ( HDL-C ) , low density lipoprotein cholesterol ( LDL-C ), malondialdehyde (MDA) , and the activities of superoxide dismutase (SOD) ,glutathione per oxidase (GSH-Px) ,induced nitric oxide synthase (iNOS) in serum were detected ,and the levels of TC ,TG, the activities of SOD,GSH-Px and induced nitric oxide synthase (iNOS) in liver were detected, and the pathological changes in aorta were observed. Results Compared with those in model group,the serum levels of TG,TC,LDL-C and the levels of TG Tc in livers in low and high dose astaxanthin groups and combined intervention group were significantly decreased ( P 〈 0.05 or P 〈 0.01 ) , however, the serum levels of HDL-C were obviously increased ( P 〈 0.01 ). The activities of SOD, GSH-Px and the content of GSH were significantly increased ( P 〈 0.05 or P 〈 0.01 ) , but the activity of iNOS and content of MDA were obviously decreased ( P 〈 0.05 orP 〈 0.01 ). Conclusion Astaxanthin has obvious effects of reducing blood fat, and has inhibition effect on lipid peroxidation injury in arteryand liver induced by high blood fat, especially the improvement effect of high dose astaxanthin is more satisfactory.
