解淀粉芽孢杆菌BW-13产生的抗真菌物质特性研究与初步分离纯化 被引量:10

摘要 解淀粉芽孢杆菌(Bacillus amyloliquefaciens)BW-13发酵滤液对酿酒酵母(Saccharomyces cerevisiae)和植物病原真菌灰霉(Botrytis cinerea)均具有显著的抑制活性,并具有热稳定性好、对紫外线照射和蛋白酶处理不敏感、在pH 3~9范围内稳定,尤其在酸性条件下基本不丧失活性的特点.通过发酵液预处理,活性炭吸附,D293强碱性季铵型阴离子交换树脂柱层析,对解淀粉芽孢杆菌BW-13产生的抗真菌物质进行了初步分离纯化.高效液相色谱(HPLC)分析显示分离得到的抗真菌物质在220nm处有单一的活性峰. Bacillus amyloliquefaciens BW-13 has a significant inhibitory activity against S.cerevisiae and plant pathogenic fungus Botrytis cinerea.The experiments showed that the crude extract of Bacillus amyloliquefaciens BW-13 was stable to high temperature,and resistant to protease K and ultraviolet radiation.In the range of pH3~9,the crude extracts maintained active and basically without losing any activities especially in acidic conditions.By the pretreatment of fermentation broth,active charcoal adsorption and D293anjon exchange resin column chromatography,an antifungal substance was isolated from the crude extract.A single peak in the 220 nm absorption of the antifungal substance was detected by HPLC analysis.
