脓毒症大鼠PPARγ与脂联素表达水平的相关性研究 被引量:4

摘要 目的观察脓毒症大鼠过氧化物酶体增殖物激活受体γ(PPARγ)与脂联素(ADPN)表达水平的关系。方法雄性SPF级SD大鼠30只,体重量180~200g,随机分为正常对照组、脓毒症组、脓毒症+PPARγ激动剂(罗格列酮)干预组,每组10只。采用尾静脉注射内毒素(LPS)方法制备大鼠脓毒症模型,24h后取血分离外周血单核细胞(PBMC),RT-PCR和Westernblot方法检测PBMC中PPARγ基因表达水平,ELISA检测血浆中ADPN水平变化趋势。结果与正常对照组相比,脓毒症组大鼠PBMC中PPARγ表达水平和血浆中的ADPN水平明显降低,而罗格列酮(RSG)处理的脓毒症组则显著上升。结论脓毒症大鼠PPARγ基因表达与ADPN水平降低,RSG干预的脓毒症大鼠PPARγ基因表达与ADPN水平上升,两者呈正相关性;PPARγ表达水平下调可能是脓毒症大鼠血浆ADPN表达下调的机制之一。 Objective To investigate the expression of peroxisome proliferator activated receptors γ(PPARγ) and adiponectin(ADPN) during sepsis in rat,and to analyze the relationship between them.Methods Thirty male SD rats of 180~200 g were randomly divided into three groups,the control group,sepsis group,and sepsis+PPARγ agonist group(rosiglitazone).Experimental models of rats with sepsis were constructed by injection with lipopolysaccharide(LPS) through caudal vein.The peripheral blood monouclear cells(PBMC) of rats were separated after 24 h.The mRNA of PPAR-γ was detected by real-time PCR,the protein levels of PPARγ and ADPN were detected by Western-blot and ELISA.Results Compared with the control group,the expression of PPARγ and ADPN were significantly decreased during sepsis in the sepsis group,while that in the sepsis+PPARγ agonist group was significantly increased.Conclusion Under the intervention of RSG,the expression of PPARγ and ADPN were significantly increased during sepsis in rat.The expression of ADPN was positively related to the expression of PPARγ.
