“通腑”方法对直肠半结扎模型大鼠肺组织NKA、VIP、TFF3含量的影响 被引量:6

Influences of therapy of relaxing bowels on content of lung NKA,VIP and TFF3 in rat model of rectal semi-ligation

摘要 目的观察直肠半结扎模型大鼠肺组织中血管活性肠肽(VIP)、神经激肽A(NKA)及肠三叶因子(TFF3)含量变化及通腑中药(大承气汤)对其的调节作用,探讨"肺合大肠"神经肽机制。方法制备直肠半结扎大鼠模型。50只清洁级SD大鼠随机分为正常组、持续结扎组、解扎组、解扎给药组、正常给药组,给药组灌胃大承气汤3 g/只。检测各组大鼠支气管肺泡灌洗液(BALF)中肿瘤坏死因子(TNF-α)、血清白细胞介素-1β(IL-1β)及肺组织中VIP、TFF3、NKA的含量。结果模型组大鼠BALF中TNF-α、IL-1β及肺组织中NKA、TFF3含量均明显升高(P<0.01),VIP含量明显降低(P<0.01)。经大承气汤从肠干预后,BALF中TNF-α、IL-1β及肺组织中NKA、TFF3含量均明显降低(P<0.01),VIP含量明显升高(P<0.01)。Pearson correlation分析显示肺组织中NKA、VIP、TFF3含量与BALF中TNF-α、IL-1β含量明显相关(P<0.01或P<0.05)。结论通腑疗法可减轻直肠半结扎模型大鼠的肺部炎症,这一效应的产生与肺组织中VIP、TFF3、NKA含量的变化密切相关。 Objective To observe the content changes of lung vasoactive intestinal peptide (VIP), neurokinin A (NKA) and intestinal trefoil factor 3 ( TFF3 ) and regulation effect of Chinese medications with action of relaxing bowels (Da Chengqi Tang), and discuss the neuropeptide mechanism of lung associating with large intestine. Methods The rat model of rectal semi-ligation was established and 50 SD rats were randomly divided into normal group, sustained ligation group, de-ligation group, de-ligation and medicated group and normal medicated group. The content of tumor necrosis factor-α (TNF-α) and interleukin-1 β (IL-1β) in bronchoalveolar lavage fluid (BALF) and VIP, TFF3 and NKA in lung tissue were detected in rats of all groups. Results The content of TNF-α and IL-1 β in BALF and NKA and TFF3 in lung tissue increased significantly (P 〈 0.01 ) and VIP decreased significantly ( P 〈 0.01 ) in sustained ligation group and de-ligation group. After the intervention with Da Chengqi Tang in intestine, the content of TNF-α and IL-1 β in BALF and NKA and TFF3 in lung tissue decreased significantly ( P 〈 0.01 ) and VIP increased significantly (P 〈0.01 ). Pearson correlation analysis showed that the content of NKA, VII) and TFF3 in lung tissue were significantly correlated to the content of TNF-α and IL-1α in BALF (P 〈 0.01 or P 〈 0.05 ). Conclusion The therapy of relaxing bowels can relieve pulmonary inflammation in rat model of rectal semi-ligation, and this effect is closely related to the content changes of VIP, TFF3 and NKA in lung tissue.
