脂多糖受体CD14在急性脑出血应激性肝损害中的作用及星蒌承气汤的效应研究 被引量:3

摘要 目的研究白细胞分化抗原14(CD14)在急性脑出血应激性肝损害内毒素信号转导通路中的作用以及星蒌承气汤的靶向调控机制。方法采用大脑注射Ⅶ型胶原酶法建立SD大鼠急性脑出血模型,随机分为正常组、脑出血组、脑出血痰热腑实证组、星蒌承气汤组,造模后24、48、72 h实时荧光定量PCR观察肝组织CD14 mRNA表达,并观察肝组织HE染色结果。结果脑出血急性期肝组织CD14 mRNA表达升高,星蒌承气汤能降低急性脑出血痰热腑实证肝组织CD14 mRNA表达,并能明显减轻肝组织病理损伤。结论 CD14在介导内毒素性肝损伤中起重要作用,星蒌承气汤对脑出血急性期痰热腑实证肝脏具有一定的保护作用。 Objective To study the role of cluster differentiation antigen 14 (CD14) in signal transduction pathway of endotoxin in hepatic stress injury of acute cerebral hemorrhage and target regulation mechanism of Xinglou Chengqi Tang. Methods The model of acute cerebral hemorrhage was established by using brain injection of eollagenase VII, and then the rats were divided into normal group, cerebral hemorrhage group, group of phlegm-heat and bowel-excess syndrome and Xinglou Chengqi Tang group. The expression of CD14 mRNA and HE staining of liver tissue were observed by applying real-time fluorescence quantitative polymerase chain reaction (RT-PCR) after modeling for 24, 48 and 72 hours. Results The expression of CD14 mRNA increased in acute stage of cerebral hemorrhage, while Xinglou Chengqi Tang reduced the expression and relieve the pathological injury of liver tissue in syndrome of phlegm heat and bowel excess of acute cerebral hemorrhage. Conclusion in mediating endotoxie liver injury, and Xinglou Chengqi Tang has some CD14 plays an important role orotective effect on liver in syndrome of phlegm heat and bowel excess of acute cerebral hemorrhage.
