丹参多酚酸对大鼠心肌线粒体呼吸链酶复合体活性的影响 被引量:7

The effect of Salvianolic acid on activity of respiratory chain enzyme complex in myocardial mitochondria of rats

摘要 目的探讨丹参多酚酸对大鼠离体心肌缺血/再灌注(I/R)损伤的作用。方法采用缺血30min、再灌注60min复制I/R损伤模型。应用紫外分光光度仪检测对照组、I/R组和丹参多酚酸组(n=8)间大鼠心肌线粒体呼吸链酶复合体Ⅰ-Ⅳ的活性。结果I/R组心肌线粒体呼吸链酶复合体Ⅰ-Ⅳ活性明显低于对照组(复合体Ⅰ:4.49±0.30比22.94±2.14;复合体Ⅱ:5.34±0.81比15.80±1.07;复合体Ⅲ:5.40±0.54比15.67±2.90;复合体Ⅳ:10.19±2.29比26.08±2.23,均P〈0.05);丹参多酚酸组I~Ⅳ活性(分别为16.12±0.43、7.72±0.55、10.37±0.58、14.49±0.86)明显高于I/R组,但仍低于对照组(均P〈0.05)。结论丹参多酚酸能够相对增加线粒体呼吸链酶复合体Ⅰ~Ⅳ的活性,改善其能量代谢,减少了氧自由基的生成,实现对I/R损伤心肌的保护作用。 Objective To investigate the effect of Salvianolic acid on myocardial ischemia/reperfusion ( I/R ) injury to isolated rat heart. Methods The model of myocardial damage of rats was made by ischemia 30 minutes and reperfusion 60 minutes. The activities of myocardial mitochondrial respiratory chain enzyme complex I-IV of control group, I/R group and Salvianolic acid-treated I/R group ( each, n=8 ) were determined by ultraviolet spectrophotometry. Results The activities of myocardial mitochondrial respiratory chain enzyme complex I -IV in 1/R group was less than that in control group ( complex 1:4.49 ± 0.30 vs. 22.94 ± 2.14; complex II: 5.34± 0.81 vs. 15.80± 1.07; complexIII : 5.40±0.54 vs. 15.67 ±2.90; complex IV: 10.19 ±2.29 vs. 26.08±2.23, all P〈0.05 ) ; The activities of myocardial mitochondrial respiratory chain enzyme complex I-IV in Salvianolic acid-treated I/R group ( 16.12 ± 0.43, 7.72± 0.55, 10.37 ± 0.58, 14.49± 0.86 respectively ) were obviously higher than those in I/R group, but still lower than those in control group (all P〈0.05). Conclusion Salvianolic acid has a protective effect on myocardial I/R injury by increasing the activities of myocardial mitochondrial respiratory chain enzyme complex I ~IV, decreasing the production of oxygen free radical and improving the metabolism of energy.
