摘要
探讨蓝舌病毒湖北株3(BTV-HbC3)对动物在体肿瘤的治疗价值及蓝舌病毒湖北株3是否增加放射治疗的疗效。建立C57BL/6小鼠前列腺癌荷瘤动物模型,设空白对照组、BTV-HbC3治疗组、放疗组和BTV-HbC3联合放疗组,观察并绘制肿瘤生长曲线图,肿瘤组织行常规HE病理学检测。结果全部小鼠均成功荷瘤。治疗后BTV-HbC3联合放疗组肿瘤体积明显小于其他各组,肿瘤体积增长时间延长,HE染色观察BTV-HbC3联合放疗组肿瘤坏死面积增加。显示BTV-HbC3联合放疗存在显著的协同效应。BTV-HbC3联合放疗对小鼠前列腺癌肿瘤生长具有显著抑制作用并可介导凋亡,为前列腺癌进一步的放疗基因治疗研究奠定了基础。
This study was designed to evaluate the treatment value of bluetongue virus strain HbC3 (BTV--HbC3) for animal tumor in vivo and investigate radiosensitive effects of BTV-- HbC3. Subcutaneous tumor -- bearing mice models of prostate cancer were created up successfully using the C57BL/6 mice and the mice were randomly divided into control group,BTV-- HbC3 group, radiotherapy group,BTV- HbC3 combined with radiotherapy group. Tumor growth curve was drawn,and HE pathological examination was performed. RM--1 prostate tumor model was successfully established in all the mice. The average gross tumor volumes in the BTV-- HbC3 combined with radiotherapy group was significantly reduced compared with other groups after treatment, tumor's doubling time prolonged, Histological examination showed the signs of tumor cell injury in BTV--HbC3 combined with radiotherapy group,there was notable synergistic effects between BTV--HbC3 and radiotherapy. Bluetongue virus strain HbC3(BTV--HbC3) combined with radiotherapy had significant inhibition to the tumor --bearing mice models of prostate cancer and induced cell. This provided the foundation for further gene radiation therapy of breast cancer.
