摘要
目的 体外观察含HCV高变区1多抗原肽(MAP)及其抗血清对HCV感染有无阻断作用。 方法 将前期合成的MAP及其抗血清和HCV感染血清按不同比例充分混合并在37℃下孵育2h或4℃下孵育12h,再将此混合血清接种7721细胞,分别以逆转录-聚合酶链反应、免疫组化、原位杂交检测细胞的感染情况。 结果 各比例混合的阻断血清组在孵育后细胞和培养上清中均可检出HCV RNA;免疫组化证实有表达HCV抗原的阳性细胞。 结论 前期合成的HCV包膜糖蛋白及其抗血清在体外不能明确阻断HCV感染/吸附肝细胞。
AIM To study whether hyperimmune rabbit serum againstMAP containing HVR1 sequences could block in vitroinfection of HCV.METHODS The hyperimmune rabbit serum was mixedwith the infectious serum of HCV according to a series ofproportion and then the mixture was incubated for 2 hoursat 37℃ or for 12 hours at 4℃. Then 7721 cells wereincubated in the mixed sera. After incubation, thepresence of HCV RNA and antigen in cells and/orsupernatant was examined by RT-PCR,immunohistochemistry and in situ hybridizationrespectively.RESULTS HCV RNA and NS3 antigen could be detected in7721 cells and/or supernatant in all blocking groups afterincubation.CONCLUSION MAP and the hyperimmune serum againstMAP can not block viral attachment or infection to ceils.
