摘要
目的:研究哮喘豚鼠肺组织G蛋白α亚基含量的表达变化及胸腺素对其的影响。方法:用卵蛋白雾化吸入致敏的方法复制豚鼠哮喘模型,并用胸腺素治疗,Western blot杂交分析法测定Gα含量。结果:哮喘组豚鼠肺组织Giα(126% ±11% )、Gqα(122% ±12% )较正常对照组显著升高(P均< 0.05);而胸腺素组Giα、Gqα(分别为97% ±9% 和98% ±11% )较哮喘组显著降低(P均< 0.05);胸腺素组Gsα(107% ±6% 和104% ±5% )无显著变化(P均> 0.05)。结论:哮喘肺组织Giα、Gqα表达增加在哮喘发病中可能占有一定地位,而胸腺素通过降低Giα、Gqα在哮喘豚鼠肺组织的表达变化达到了治疗或缓解哮喘的目的。
Objective:To study the alteration in Gproteins in tissues in asthmatic guinea pigs.Methods:Guinea pigs were sensitized and challenged with ovalbumin and treated with thymosin.The levels of Gsα,Giα,Goα and Gqα in lung tissue were measured with Western blot analysis.Results:The levels of Giα and Gqα in asthmatic animals were increased significantly as compared with those in group NS (126%±11% and 122%±12% vs.100%,both P <0 05).The contents of Giα and Gqα in thymosin treated animals were significantly lower than those in asthmatic ones (97%±9% and 98%+11% vs. 126%±11% and 122%±12%).There was no significant change in the level of Gsα in both groups (both P >0 05).Conclusions:The changes in Gproteins and their subfamilies,such as Gqα and Giα in lungs may be involved in the development of asthma.Thymosin could be used to treat the asthma by influencing the level of Gproteins.
