摘要
聚丙烯酰胺凝胶电泳图谱显示出,瓜-棉蚜(Aphis gossypii Glov.)种群中大多数个体都具有6条基本的酯酶活性带(E_2—E_7),但是,在东北旺抗性种群中发现了具有9条带的个体,其中E_1染色最浅,E_8、E_9染色较深。由于该蚜对拟除虫菊酯的抗性水平与其酯酶活性密切相关,确定每毫克蛋白质15分钟水解α-乙酸萘酯大于40微摩尔的蚜虫个体频率在种群中大于0.11(11%)时为抗性种群。由于AChE变构是瓜-棉蚜对有机磷和氨基甲酸酯的主要抗性机制之一,作者提出一种检测具有AChE变构抗性基因个体的方法,如果这类个体在种群中的比例大于1%,即为抗性种群。
The electropherogram of melon-cotton aphid,Aphis gossypii Glov.,obtained from polyacrylamide gel electrophoresis of single aphid of dif-ferent colonies collected from several hosts (cucumber,cotton,etc.) inBeijing suburbs,showed that all of the individuals had six esterase ac-tivity bands,E_2—E_7,while some individuals of resistant colonies hadanother three additional bands,in which E_1 stained weakly and theother two,E_8 and E_9 were more intense.Whether E_8 and E,conferedresistance needs further investigation.The resistant levels of differentcolonies of melon-cotton aphids to fenvalerate were significantly corre-lated with the esterase activity of individual demonstrated before,wepropose that the colonies in which frequency of the individuals whoseesterase activities are more than 40 μmol/mg protein,fifteen min basedon hydrolysis of 1-NA is more than 11 percent are resistant populations.A method for detecting the altered acetylcholinesterase resistancegenes,which confered organophosphate and carbamate insecticides re-sistance,in single melon-cotton aphid was developed.Because the in-sensitivity of AChE was the major resistant mechanism of the melon-cotton aphid,which had been searched before,we also propose that thecolonies in which the proportion of the individuals with altered AChEresistance genes is more than one percent are resistance populations.
