摘要
目的:探讨碱性成纤维细胞生长因子(basic fibroblast growth factor ,bFGF) 对关节软骨细胞创伤愈合与增殖行为的影响。方法:采用体外创伤愈合模型及MTT 比色方法,对软骨细胞创伤愈合与增殖行为进行分析。结果:bFGF浓度为5ng/ ml 时,即可显著促进软骨细胞创伤愈合,浓度为50ng/ ml 时,其促进作用达到最大值。bFGF 也可促进软骨细胞增殖,1ng/ ml bFGF 即可促进第2 代软骨细胞增殖,但不影响第5 代细胞的增殖行为,当bFGF 达到10ng/ ml 时,才显著地促进第5 代细胞增殖。bFGF 浓度高至100ng/ ml,其对两代细胞的促进作用不再提高。结论:bFGF 能够促进体外软骨细胞的创伤愈合和增殖,但对不同代数细胞的增殖作用有所不同,bFGF 促进细胞创伤愈合可能与其促进增殖有关。
Objective: To study the effects of basic fibroblast growth factor(bFGF) on wound healing and proliferation of chondrocytes.Methods:The wound healing and proliferation of chondrocytes were assayed in a healing model in vitro and by MTT colorimetric method respectively.Results:bFGF significantly enhanced the wound healing of chondrocytes at the concentration of 5ng/ml,and this enhancement was maximal in the presence of 50ng/ml bFGF.bFGF also enhanced the proliferation of chondrocytes.1ng/ml bFGF significantly increased the proliferation of chondrocytes in the second passage,but had no effect in the fifth passage until the bFGF concentration got to 10ng/ml.Increasing the bFGF to 100ng/ml did not show additional improvement of proliferation in both passages.Conclusions:bFGF can enhance wound healing and proliferation of chondrocytes in vitro,but the proliferation response of chondrocytes in different passages may be different,the enhancement of wound healing by bFGF may be related to the cell proliferation response.
