善得定对门静脉高压性胃病大鼠胃粘膜灌注的影响 被引量:20

Effects of sandostatin on gastric mucosal perfusion in rats with portal hypertensive gastropathy

摘要 目的观察善得定对门静脉高压性胃病(portal hypertensive gnstropathy, PHG)大鼠胃粘膜血流量(gastric  mucosal blood flow, GMBF)的影响,并对其作用机制作初步探讨。方法部分结扎大鼠门静脉主干2周后,观察善得定对 PHG大鼠全身血流动力学、GMBF、门静脉压力(PVP)的影响,测定了输往善得定 30 min后 PHG大鼠血浆胰高糖素、血浆和胃粘膜NO2-/NO3、6-酮-前列腺素F1a(6-Keto-PGF1a)含量,并与心得安进行对比。结果善得定和心得安均不能拮抗前列环素,能降低 GMBF、 PVP;善得定组与 PHG对照组血浆胰高糖素分别为(220.15 ± 8.12) pg/ml和(329.6718.62)pg/ml;血浆NO2-/NO3-分别为(70.30±1.36)μmol/L和(80.25±1.36)μmol/L,善得定能拮抗PHG大鼠胰高糖素、NO,对平均动脉压(MAP)、心率无明显影响,心得安则与善得定相反。结论善得定通过拮抗PHG大鼠胰高糖素、NO作用降低GMBF,且对外周循环无明显影响。 Objective To observe the effects of sandostatin on gastric mucosal blood flow(GMBF) in rats with (portal hypertensive gastropathy, PHG) and its mechanisms. Methods Two weeks after portal vein ligation, the changes of systemic hemodynamics, GMBF, (portal vein pressure, PVP) were observed and plasma glucagon, plasma and gastric mucosal NO2-/CO3 -, 6 - Keto- PGF 1 a were me asured after intravenons sandostati n, propranol of and norm al saline. Results GMBF and PVP were reduced after intravenous sandostatin or propranolol. Sadostatin decreased plasma glucagon, plasma and gastric mucosal NO2-I/NO3, and had no effect on MAP and heart rate of PHG rats as compared with control group of PHG. Propranolol had not these effects, but it might decrease MAP and heart rate. There were no differences of plasma and gastric mucosal 6-Keto-PGF1a. between all PHG groups. Conclusion Sandostatin reduced GMBF of rats with PHG by hindering glucagon, NO, and had no effect on systemic hemodynamics.
