成骨生长肽对成骨细胞样细胞的成骨影响 被引量:14

Effects of osteogenic growth peptide on bone form ation of rabbit osteoblast - like cell in vitro

摘要 目的 成骨生长肽(osteogenic growth peptide , OGP) 是新近发现的具有促成骨作用和刺激造血的多肽类生长因子。本研究旨在观察成骨生长肽对兔成骨细胞样细胞的促成骨作用。方法 兔颅盖骨成骨细胞样细胞分成实验组和对照组培养, 实验组加成骨生长肽。分别测定细胞的Ⅰ型胶原m RNA 表达、碱性磷酸酶活性、胶原和钙含量。结果 培养7 d 时实验组细胞的Ⅰ型胶原m RNA 高度表达,而对照组细胞的Ⅰ型胶原m RNA 表达相对较低。培养7、10 d ,2 个时间点实验组细胞胶原和钙含量与相应的对照组比较, 差异均有非常显著性意义( P< 0-001) ,尤为显著的是实验组细胞的钙含量约为对照组的3 倍。培养3 、7、10 、14 、21 d ,5 个时间点实验组细胞的碱性磷酸酶活性与相应的对照组比较, 差异均有非常显著性意义( P< 0-01~0-001) , 有的实验组细胞的碱性磷酸酶活性约为对照组的3 ~4 倍。结论 成骨生长肽能增加细胞的Ⅰ型胶原m RNA 表达,增加碱性磷酸酶活性、胶原合成和钙盐沉积。因此,成骨生长肽调节成骨细胞样细胞的功能,使细胞分化。 Objective To investigate the effect of osteogenic growth peptide on bone formation of rabbit osteoblast - like cells cultured in vitro- Methods Osteoblast - like cells were harvested from rabbit calvariu m and divided into the control and experim ental groups- In the experimental group , an a mount of osteogenic growth peptide prepared with the method described by Greenberg was added to Os teoblast -like cells cultured in vitro- Type Icollagen m RNA expression , and alkaline phosphatase activi ty , as well as collagen and calciu m contents of osteoblast -like cells were determined in both groups- Results Type Ⅰcollagen m RNA expression level of the osteoblast -like cells increased in the experimen tal group at 7th day of the mixed culturing , ho wever , the level was lower in the control group- The Collagen and calciu m contents of the cultured cells were significantly higher in the experimental group than those in the control group on 7th and 10th days of the culturing( P< 0-001) - The Calcium con tent in the experimental group was three times as many as that in the control group- The alkaline phos phatase activity of the osteoblast like cells was significantly higher in the experim ental than in the con trol group on the third , 7th , 10th , 14th and 21st day of the culture ( P< 0-01 ~0-001)- The alkaline phosphatase activity in the experimental group was three to four times as high as that of the control group ,especially at 7th day- Conclusion Osteogenic growth peptide can increase Type I collagen m R NA expression and collagen synthesis ,and potently enhance alkaline phosphatase activity and mineraliza tion in the osteoblast -like cells- Therefore , osteogenic growth peptide plays an important role in regulat ing function , pro moting differentiation and increasing osteogenic effect of osteoblast like cells-
