多重PCR检测急性非淋巴细胞白血病IgH及TCRVγΙ-Jγ基因重排 被引量:3

Detection of IgH and TCRVγI-Jγ gene rearrangement in acute nonlymphoblastic leukemia by multiplex PCR amplification

摘要 目的:为进一步了解急性非淋巴细胞白血病(ANLL)病人基因重排情况。方法:应用多重PCR技术检测41例ANLL病人IgH及TCRVγIJγ基因重排。结果:26.8%(11/41)ANLL病人存在IgH或/和TCRVγIJγ基因重排,3例病人同时存在IgH和TCRVγIJγ基因重排;基因重排阳性ANLL治疗缓解率低于重排阴性ANLL病人(P<0.05)。结论:①IgH或TCR基因重排并非局限于淋巴细胞白血病,也可发生于部分ANLL病人;②多重PCR技术可以一次PCR扩增同时检测两种重排基因,更适于临床推广应用。 Objective:In order to get more insight into the gene rearrangement in acute non lymphoblastic leukemia(ANLL).Methods:Multiplex PCR was used to detect IgH and TCRVγI Jγ gene rearrangement in 41 ANLL cases. Results:26.8% (11/41) cases were found to have IgH or TCRVγI Jγ gene rearrangement, 3 cases were found with IgH or TCRVγI Jγ gene rearrangement ,3 cases were found with IgH or TCRVγI Jγ gene rearrangement.The complete remission rate were lower in ANLL with gene rearrangement than that in ANLL without gene rearrangement.The complete remission rate were lower in ANLL with gene rearrangement than that in ANLL without gene rearrangement. Conclusion:①IgH or TCR gene rearrangement might not be limited to lymphoid leukemia, it might occured in some ANLL cases.②Multiplex PCR can detect both IgH and TCR gene rearrangemen in a single PCR amplification and is more suitable for clinical application.
