摘要
建立家兔血浆中新抗炎镇痛剂 SFZ- 47[(± ) - 3H - 1 ,2 -二氢 - 2 - ( 4 -甲基苯胺基 ) -甲基 - 1 -吡咯里嗪酮 ]对映异构体浓度测定的高效液相色谱 ( HPL C)测定法 .采用反相 HPLC测定服药后血浆中 (± ) - SFZ- 47,同时收集其流出组分 .经浓缩后 ,再用 Chiral- AGP手性色谱柱进行对映异构体的直接分离 ,达到了良好的分离效果 .实验表明 。
A new steroselective HPLC assay has been developed for the determination of the concentration of the enantiomers of SFZ 47 [3 H 1,2 dihydro 2 (4 methylphenylamino) 1 pyrrolizinone], a novel anti inflammatory and analgesic agent, in rabbit plasma After the racemate in plasma was isolated and determined using reversed phase HPLC, the HPLC fractions containing the racemate were collected, concentrated and directly separated well on a Chiral AGP ( α 1 acid glycoprotein) column The results showed that there was significant steroselectivity for SFZ 47 in rabbits.
