摘要
为了促进小麦抗白粉病基因聚合体材料在小麦育种及生产中的应用,利用与抗白粉病基因Pm21和Pm13连锁的特异标记,在含有抗白粉病基因Pm21和Pm13的杂交F4代群体中随机挑选114个单株分别进行单一和多重PCR检测。结果表明,多重与单一PCR检测结果一致,在114个供试单株中,有5株具有Pm21,74株具有Pm13的特异标记,其中4个单株同时具有Pm21和Pm13的特异标记。与抗白粉病基因Pm21和Pm13连锁的特异标记可以用于对Pm21和Pm13的多重PCR检测,更加快速地检测出含有抗病基因Pm21和Pm13的累加体,有效应用于辅助选择育种。
In order to promote the application of polymers of genes with resistance to powdery mildew in wheat,PCR and Multiplex-PCR of 114 individuals that selected randomly from the offspring of the cross combination between Pm21 and Pm13,with the specific molecular marker linked to powdery mildew resistant gene Pm21 and Pm13,respectively.The result showed that the outcome of PCR and Multiplex-PCR was consistent,five individuals of the 114 conceive Pm21,74 conceive Pm13,and four conceive Pm21 and Pm13 simultaneously.Consequently,the specific molecular marker linked to powdery mildew resistant gene Pm21 and Pm13 in the present study could be used in Multiplex-PCR of Pm21 and Pm13,detecting the polymer of the resistant gene Pm21 and Pm13 and for better application of maker-assisted selection breeding.
