摘要
目的:研究慢性不可预见性温和应激(CUMS)对动物行为学和下丘脑胃泌素释放肽(GRP)mRNA表达的影响。方法:将14只大鼠随机分为抑郁组和对照组各7只,抑郁组采用CUMS制备抑郁模型。对2组大鼠进行行为学观察,使用荧光定量PCR检测GRP mRNA表达。结果:与对照组比较,抑郁组大鼠在应激后的糖水偏好和自主活动显著减低(P<0.001),GRP mR-NA表达升高(P<0.05)。结论:慢性应激后,出现抑郁样行为改变与GRP mRNA表达上调相关。
Objective:To investigate the changes of behavior and expression of hypothalamic gastrin-releasing peptide(GRP) mRNA after chronic and unpredictable mild stress(CUMS).Methods:Fourteen rats were randomly divided into control group(n=7) and depression group(n=7).The rats in the depression group were subjected to CUMS for 21 consecutive days.The behavior of 2 groups were observed.The expression of GRP mRNA was measured by real-time quantitative PCR.Results:The autonomic activity and sugar consumption in the depression group were significantly lower than those in the control(P0.001).The total distance,speed and rearing were significantly reduced in the stress group,revealed by the open field test(P0.001).The expression of GRP mRNA in the stress group was significantly higher than that in the control group(P0.05).Conclusion:The up-regulation of GRP mRNA probably plays a key in the behavior changes in rats exposed to stress.
