摘要
目的:探讨法舒地尔对脊髓缺血再灌注损伤的保护作用。方法:家兔30只,随机分为法舒地尔组20只和空白对照组10只,采用腹主动脉夹闭法制备脊髓缺血再灌注损伤模型,用腹腔注射法给法舒地尔组注射法舒地尔,空白对照组注射生理盐水,共14d,检测家兔血清自由基水平,治疗结束后采用Jacobs法对家兔后肢运动功能评分,评估后肢肌电图。结果:治疗后法舒地尔组家兔双后肢Jacobs评分明显高于空白对照组(P<0.001),血清丙二醛(MDA)、超氧化物岐化酶(SOD)、一氧化氮(NO)水平低于空白对照组(P<0.05或0.001),后肢运动诱发电位潜伏期(N1及P1)亦明显短于对照组(P<0.001)。结论:法舒地尔能减轻家兔脊髓缺血再灌注损伤。
Objective:To explore the neuroprotective effect of Fasudil,a Rho-kinase inhibitor,on rabbit spinal ischemia-reperfusion injury.Methods:The ischemia-reperfusion model of the spinal cord was conducted by the selective occlusion of the lumbar arteries in thirty rabbits,which were randomly divided into Fasudil(n=20) and control groups(n=10).The rabbits in the Fasudil group were given Fasudil through intraperitoneal injection whereas the control group received the equivalent physiological saline in the same way.After two weeks,the level of free ra-dical(MDA,SOD and NO) in the serum was detected,and the function of hindlimbs was evaluated by the Jacobs method.In addition,the motor evoked potentials(MEP) were also assessed.Results:Treatment with Fasudil resulted in a significant improvement of Jacobs score(P0.001),demonstrated by the decreased levels of free radicals when compared to that in the control group(P0.05 or 0.01).The MEP in the Fasudil group was shorter than that in the control group(P0.001).Conclusion:Fasudil provides a neuroprotective effect on rabbits following spinal ischemia-reperfusion injury.
