miR-200c对TGF-β_1刺激后人瘢痕疙瘩成纤维细胞胶原合成的影响

摘要 目的:观察转染mi R-200c mi mi cs对人瘢痕疙瘩成纤维细胞(HKFs)胶原蛋白分泌的影响。方法:将mi R-200c mi mi cs用ol i gof ect ami脂质体转染人瘢痕疙瘩成纤维细胞后,3H-脯氨酸掺入法测加入50μg/L剂量的转化生长因子(TGF-β1)刺激前后胶原蛋白水平的变化。结果:转染mi R-200c mi mi cs后,胶原蛋白分泌水平降为正常皮肤成纤维细胞表达水平;经TGF-β1刺激后,其蛋白水平仍无明显变化。结论:mi R-200c可明显抑制人瘢痕疙瘩成纤维细胞胶原蛋白分泌,推测其机制是通过抑制TGF-β1生物学作用。 Objective To investigate the effect of miR-200c mimics on collagen secretion in human keloid fibroblasts(HKFs).Methods miR-200c mimics were transfected by oligofectami package.The collagen level was measured by 3H-proline incorporation method before and after HKFs were stimulated by transforming growth factor betal(TGF-β1) of the dosage of 50μg/L.Results The collagen level was decreased to the normal skin fibroblasts level and had no obvious change even after stimulated by TGF-β1.Conclusion miR-200c can inhibit the expression of collagen in HKFs.The mechanisms may be inhibit the biological effects of TGF-β1.
