摘要
目的观察眼针对脑缺血再灌注模型大鼠海马组织细胞间黏附因子(intercellular adhesionmolecule,ICAM)表达的影响,探讨眼针对脑缺血再灌注损伤治疗的机制。方法应用线栓法复制脑缺血再灌注损伤大鼠模型,采用眼针治疗,于缺血再灌注后72h进行神经功能缺损评分,采用Western blot方法、实时荧光定量聚合酶链反应(RQ-PCR)方法测定海马组织ICAM蛋白及mRNA表达的变化。结果缺血再灌注损伤模型组分别与正常组和假手术组比较,大鼠神经功能缺损评分明显升高,且Western结果显示模型组ICAM蛋白量升高,mRNA的表达上调。与模型组比较,眼针组大鼠神经功能缺损评分明显下降,眼针组大鼠海马组织ICAM蛋白及mRNA表达明显下调(<0.05)。结论眼针具有明显的改善大鼠脑缺血再灌注损伤的作用;其机制之一可能与下调海马组织ICAM表达有关。
Objective To investigate the effect of eye acupuncture therapy on ICAM expression in hippocampus of rats with acute cerebral ischemia-reperfusion injury.Methods To establish the rat model of cerebral ischemia-reperfusion by suture method,the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks after ischemia-reperfusion 72h.Western-blotting and RQ-PCR were taken to detect the alteration of ICAM protein and mRNA expression in rat hippocampus.Results Compared with the normal group and sham group,the neurologic impairment score and the expression of the ICAM protein and mRNA in the ischemia-reperfusion model group were both increased obviously.Compared with the model group,the neurologic impairment score and the expression of ICAM protein and ICAM mRNA in rat hippocampus in eye acupuncture therapy group were decreased obviously(0.05).Conclusions The improvement of cerebral ischemia-reperfusion injury by eye acupuncture therapy might be related to the downregulation of the ICAM protein expression in hippocampus.
