摘要
目的观察雌激素(E2)对乳腺癌MCF7细胞中E-钙粘蛋白基因(CDH)的调控。方法使用Western blot、Real Time PCR分别在蛋白和RNA水平上观察雌激素如何影响E-钙粘蛋白。使用双荧光素酶报告系统检测雌激素如何影响E-钙粘蛋白启动子活性。结果雌激素下调乳腺癌MCF7细胞中E-钙粘蛋白的蛋白和RNA水平,并且能够下调E-钙粘蛋白的启动子活性。结论雌激素下调乳腺癌细胞中E-钙粘蛋白基因,为进一步探讨乳腺癌发病机制提供有力证据。
Objective To observe how β-Eatradiol regulates E-cadherin gene in breast cancer MCF7 cell line.Methods Western blot was used to detect E-cadherin expression in the absence or presence of β-Eatradiol.Real Time PCR was used to detect the expression of E-cadherin RNA in the β-Eatradiol treatment,dual luciferase report system was used to assay E-cadherin promoter activity in the same condition.Results β-Eatradiol downregulates expression of E-cadherin protein,mRNA and activity of E-cadherin promoter in breast cancer MCF7 cell line.Conclusion Downregulation of E-cadherin gene by β-Eatradiol would provide potent proof for researching breast cancer pathogenesis.
