人骨髓间充质干细胞对体外培养的胎儿神经干细胞向少突胶质细胞分化的影响 被引量:1

Effects of MSCs on differentiation of fetus NSCs into oligodendrocytes in vitro

摘要 目的探讨骨髓间充质干细胞(MSCs)对体外培养的神经干细胞(NSCs)向少突胶质细胞分化的影响。方法以密度梯度离心的方法获得人hMSCs,设立3个实验组,分别是hMSCs+NSCs共培养组、hMSCs条件培养基+NSCs组、NSCs自发分化组(对照组)。通过免疫细胞化学染色及RT-PCR等手段检测不同组中NSCs向少突胶质细胞分化的情况,并探讨其可能的机制。结果与自发分化组相比,共培养组及hMSCs条件培养基作用组NSCs向少突胶质细胞分化的百分率分别为(61.3±5.7)%和(58.4±6.2)%,差异均具有显著性(<0.05);且RT-PCR结果显示,共培养组及hMSCs条件培养基作用组少突胶质细胞相关基因Olig1、Olig2的表达增强。结论 hMSCs促进体外培养的NSCs向少突胶质细胞分化,可能与hMSCs分泌可溶性因子相关。 Objective To investigate the effect of human bone mesenchymal stem cells(hMSCs)on differentiation of neural stem cells(NSCs) into oligodendrocytes in vitro.Methods hMSCs and NSCs were obtained by density gradient centrifugation and by suspension neurospheres respectively,the experiments were divided into 3 groups:hMSCs +NSCs co-culture group,hMSCs conditioned media(CM)+NSCs group and NSCs group.We observed the effect of hMSCs on differentiation of NSCs by immunofluorescence staining and RT-PCR.Results Compared with NSCs group,the percentage of oligodendrocytes increased remarkably in hMSCs+NSCs coculture group(61.3±5.7)%and hMSCs+CM group(58.4±6.2)%respectively(0.05),the expression levels of genes Olig1 and Olig2 were enhanced in hMSCs+NSCs coculture group and hMSCs+CM group by RT-PCR.Conclusion The promotion of hMSCs on NSCs differentiation into oligodendrocytes might be related to activation of the secreted soluble factors by hMSCs.
