超声处理对人参种子萌发以及SOD、POD的影响 被引量:6

摘要 目的:人参种子具有双重休眠的特点,要经过一定时间的形态后熟与生理后熟才能萌发。影响种子萌发内在条件是胚的完整性,其内在的酶系统及生化反应也是影响种子发芽的重要生理指标。其中SOD(超氧化物歧化酶)、POD(过氧化物酶)直接影响种子活力。方法:通过超声波处理来探索对人参种子萌发的影响。结果:超声温度50℃时,裂口率为90%,高于CK13百分点;时间30min时,裂口率为90%,高于CK13百分点;功率550W时,裂口率为95%,高于CK18百分点。而超声温度为30℃时,胚率是66.6%,高于CK6.0百分点;时间为20min时,胚率是65.3%,高于CK4.7百分点;功率为450W时胚率是49.7%,低于CK10.9百分点。通过方差分析,PSOD=0.0001<0.001,说明超声处理对种子SOD有很高的显著性;PPOD=0.0001<0.001,说明超声对种子POD的影响极显著。结论:可以作为生产上采用超声波处理人参种子的理论依据。 Objective: The Ginseng seed has the character of double dormancy, when the seeds finished morphological after-ripening and physiological after-ripening, it will germinate. Dormancy of seeds had a relationship with It's integrity. The enzyme system of seeds is one of important impacts on seed germination physiological parameters. The SOD and POD can impact on seed vigor directly. Methods: In this experiment, ultrasonic method was applied to explore, that how to impact on seed germination. Results: when the temperature is 50 ℃, the rate gap of seed can reach 90 %. When the time is 30 min, the rate gap of seed is 90 %. Double of them are higher than CK is 13 percentage points. When the power is 550 W, the rate gap of seed is 95 %. It is higher than CK 18 percentage points. When the temperature is 30 ℃, the embryo rate of seed is 66. 6 %. When the time is 20 min, the embryo rate of seed is 65.3 %. When the power is 450 W, the embryo rate of seed is 49.7 %. The embryo rate of CK is 60. 6 %. Analysis of variance, PSOD = 0. 000 1 〈 0. 001, so we can see that the ultrasonic method could affect on SOD very extraordinary. And PPOD = 0. 000 1 〈 0. 001, so the affect on SOD is also extraordinary. Conclusion: It could be as the theoretical basis of using ultrasonic on Ginseng seed in practice.
