摘要
目的建立HPLC法测定头孢唑肟钠氯化钠即配型输液中头孢唑肟含量的方法。方法选用C18柱,以pH 3.6缓冲液(取枸橼酸1.42 g、磷酸氢二钠2.31 g,加水溶解并稀释至1 000 ml)-乙腈(9∶1)为流动相,流速1.0 ml/min,进样量20μl,检测波长254 nm,柱温25℃。结果头孢唑肟线性范围为60.04-140.09μg/ml,回归方程为Y=20 920 X+31 143(r=0.999 9);平均回收率99.69%,RSD=0.20%。结论该HPLC法可用于头孢唑肟钠氯化钠即配型输液中头孢唑肟的含量测定。
Objective To establish a high performance liquid chromatography(HPLC) method for determining the content of ceftizoxime in ceftizoxime sodium for injection / sodium chloride injection.Methods HPLC was performed using a C18 column with mixed solvent of pH 3.6 buffer solution(dissolving 1.42 g of citric acid monohydrate and 2.31 g of disodium hydrogen phosphate into water to obtain 1 000 ml of solution)-acetonitrile(9∶ 1) as the mobile phase at the flow rate of 1.0 ml/min.The injection volume was 20 μl,the wavelength was set at 254 nm and the column temperature was 25 ℃.Results The ceftizoxime showed a good linear relationship between the concentration and peak area in the range of 60.04-140.09 μg/ml,and the regression equation was Y=20 920X+31 143(r=0.999 9).The average recovery was 99.69%,and RSD was 0.20%.Conclusion The established HPLC method can be used to determine the content of ceftizoxime in ceftizoxime sodium for injection / sodium chloride injection.
